Methods and systems for extracting nucleic acids from a biological sample
Summary
The USPTO granted patent US12595475B2 to Kryptos Biotechnologies, Inc. on April 7, 2026, covering methods and systems for extracting nucleic acids from biological samples using solid-phase substrate binding and magnetic field trapping at a chamber site. The patent includes 11 claims for an automated purification process involving wash buffer, immiscible fluid, and elution buffer steps.
What changed
The USPTO has formally granted patent US12595475B2 to Kryptos Biotechnologies, Inc. for methods and systems involving nucleic acid extraction using solid-phase substrate capture and magnetic trapping technology. The B2 kind code indicates this is a granted patent following examination. The patent protects a fluidic process where nucleic acids bind to a solid substrate, are magnetically trapped, washed with buffer solutions, treated with immiscible fluid to remove residuals, and eluted for downstream use.
This patent grant provides Kryptos Biotechnologies, Inc. exclusive rights to practice their magnetic field-based nucleic acid extraction method in the United States until the patent expires. Competitors developing similar automated nucleic acid purification systems should assess whether their methods fall within the scope of the 11 granted claims. Research institutions and diagnostic kit manufacturers may need to consider licensing this technology or designing around the claims.
What to do next
- Record the patent grant and expiration date
- Monitor for potential infringement of the patented method
- Review product development activities for freedom-to-operate implications

Abstract
A method for extracting nucleic acids includes mixing a biological sample with a solid-phase substrate to produce a sample fluid. The nucleic acids in the sample fluid bind to the solid-phase substrate. The method also includes flowing the sample fluid in a fluid conduit to a trapping site. The trapping site may include a chamber. The method may further include applying a magnetic field to trap the solid-phase substrate of the sample fluid flowing through the fluid conduit at the trapping site. The method further includes flowing a wash buffer through the fluid conduit to remove impurities from the solid-phase substrate. The method further includes flowing an immiscible fluid through the fluid conduit to remove residual sample fluid and/or wash buffer. The method further includes flowing an elution buffer through the fluid conduit to elute nucleic acids from the solid-phase substrate.
